Kit utilizes Live-dye™, a cell permeable green fluorescent dye (Ex(max): 488nm; Em(max): 518nm), to stain live cells. Dead cells can be easily stained by propidium iodide (PI), a red fluorescent dye (Ex(max): 488nm; Em(max): 615nm), which in viable cells is actively pumped out of the cytoplasm. Stained cells can be visualized by fluorescence microscopy using a band-pass filter (detects FITC and rhodamine). RBN-2397 selectively inhibits PARP-7 and demonstrates >50-fold selectivity for inhibition of PARP-7 over all PARP family members as measured by biochemical assays. RBN-2397 inhibits PARP-7-dependent MARylation IC50 = 2nM in cell biochemical assay and inhibits cells proliferation in NCI-H1373 lung cancer cells IC50 = 20nM. By selectively inhibiting PARP-7 in tumor cells, RBN-2397 has been shown to directly inhibit cellular proliferation and restore Type I interferon signaling to stimulate an innate and adaptive antitumor immune response. Targeting cytosolic nucleic acid sensing pathways and the Type I interferon IFN response is an emerging therapeutic strategy being explored in oncology. The PARP family consists of seventeen enzymes that regulate fundamental biological processes including response to cellular stress.

Annexin IV (ANX4) belongs to the annexin family of calcium-dependent phospholipid binding proteins. Although their functions are still not clearly defined, several members of the annexin family have been implicated in membrane-related events along exocytotic and endocytotic pathways. ANX4 has 45 to 59% identity with other members of its family and shares a similar size and exon-intron organization. Isolated from human placenta, ANX4 encodes a protein that has possible interactions with ATP, and has in vitro anticoagulant activity and also inhibits phospholipase A2 activity. ANX4 is almost exclusively expressed in epithelial cells. Several transcript variants encoding different isoforms have been found for this gene.

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 3 and 7, and the protein itself is processed by caspase 8. Mutations in this gene are associated with type IIA autoimmune lymphoproliferative syndrome, non-Hodgkin lymphoma and gastric cancer. Alternatively spliced transcript variants encoding different isoforms have been described for this gene.
